    My genetic research focuses on the mechanisms that generate and maintain variation in the fungus Microbotryum violaceum. In particular, I am interested in the interactions between reproductive biology and karyotype evolution and the distribution of heterozygous loci. The reason M. violaceum is particularly well suited for such studies is that it forms linear tetrads (figure1.jpg), which can be isolated by micromanipulation and cultured on standard growth media. This allows examination of each haploid product of a single meiosis and reconstruction of the parental diploid condition. It also allows for complete crossing designs to test the consequences of particular types of mating.

    The tetrads I've isolated may also be useful for other studies, and I've listed my current collection in the table below. The collection is available to those interested in M. violaceum for scholarly research or for teaching purposes (i.e. one nice use of M. violaceum in teaching is to demonstrate reproductive isolation among host-races by conjugation rates in vitro; more information available).

    Obtaining Samples: An approved APHIS form PPQ-526 (PDF link) is required for interstate movement of M. violaceum (similar requirement may be needed for movement between countries). It will facilitated approval if the following facts are mentioned: 1) M. violaceum does not cause disease of agriculturally important plants (lineages are particularly host-specific), 2) it is not closely related to the smut fungi found on cereal crops, 3) the host-race on Silene latifolia is naturalize in much of North America, and 4) the samples will be sent as separate haploid cultures, which are not pathogenic. There is no cost of obtaining samples beyond what would be required of time and materials, and you should send your request to me at the Department of Biology, University of Virginia, Charlottesville, VA 22903. The tetrads can be kept indefinitely when frozen under desiccation, and then restored by plating to standard growth media, like potato dextrose agar. * Deciphering the Table:  This collection also includes a few tetrads from other smut fungi.  And sorry about the extensive use of codes and abbreviations.  T, P, M, and D correspond to the position of cells in the meiotic tetrads (as in animation above), and the "x" in the table indicates which ones we have stored. In the "STORED" column, “complete” means we have isolated and stored each of the four meiotic products, “partial” means an incomplete tetrad was isolated (often the T and D cells which are of opposite mating types).  If just “A1” and/or “A2” are indicated in this column, this means the mating types of the cultures are known but not their original positions within the tetrads.
